I'm assuming TDC for leak down test, if so, is there a locking bolt or mechanism to hold the crank and keep the piston at TDC with 100 psi applied?

Nope. People just use a rod or tire iron through the primary to the fender of floor. It can be in any position where all the valves are closed..ie between 1/2 way up on the compression stroke to 1/2 way down on the power stroke.
